This Rat Serum Amyloid P Sandwich ELISA Kit from Innovative Research is intended for quantitative detection of rat SAP in cell culture supernates, cell lysates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.

This rat SAP Sandwich ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for SAP has been precoated onto 96-well plates. Standards(NSO, Q21-S228) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for SAP is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the rat SAP amount of sample captured in plate.

The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. Expression system for standard: Amyloid P component, serum(SAP), also known as PTX2 or APCS, is the identical serum form of amyloid P component(AP), a 25kDa pentameric protein first identified as the pentagonal constituent of in vivo pathological deposits called "amyloid". It belongs to the pentraxins family, characterised by calcium dependent ligand binding and distinctive flattened beta-jellyroll structure similar to that of the legume lectins. This gene is mapped to 1q23.2. The binding of the encoded protein to proteins in the pathological amyloid cross-beta fold suggests its possible role as a chaperone. This protein is also thought to control the degradation of chromatin. It has been demonstrated that this protein binds to apoptotic cells at an early stage, which raises the possibility that it is involved in dealing with apoptotic cells in vivo.; Immunogen sequence: 369



Curious About The Best Way To Thaw Plasma?


  • Preheat a water bath to 37 C.
  • Place the frozen plasma sample directly in the water bath.
  • Gently agitate the plasma sample every 5-10 minutes to insure even thawing.
  • The total thawing time will depend on the frozen sample volume. A 10 ml sample might be thawed completely in 5 minutes, but a large sample could take up to 1 hour.
  • The thawed sample should be used immediately or stored at 4 C for up to 24 hours. Liquid plasma should never be kept on ice or stored below 0 C as the cold may cause clotting.
  • Plasma should be visually inspected for signs of clotting or precipitation before use in experiments. A coagulation test such as aPTT can also be performed to verify the preservation of the hemostasis system.
  • Plasma should always be snap-frozen by placing the tube in dry ice for at least 1 hour. Large containers can be covered in dry ice for even freezing.
  • Frozen plasma should be stored at –20 C or colder. Lower temperatures will help preserve labile plasma proteins for longer time periods.

This protocol is applicable for plasma containing any anticoagulant of both human and animal origin. While handling serum does not involve the same risk of clotting due to improper handling, we recommend following the above instructions for both plasma and serum for best results
