This rat MAG ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for MAG has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: G20-P516) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for MAG is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the rat MAG amount of sample captured in plate.

Expression system for standard: Myelin-associated glycoprotein (MAG, Siglec-4) is a cell membrane glycoprotein that is a member of the SIGLEC family of proteins and is a functional ligand of the NoGo-66 receptor(NgR). This gene is mapped to 1q21. The protein encoded by this gene is a type I membrane protein and member of the immunoglobulin superfamily. It is thought to be involved in the process of myelination. It is a lectin that binds to sialylated glycoconjugates and mediates certain myelin-neuron cell-cell interactions.
