This Rat Endothelial Leukocyte Adhesion Molecule 1 (E-Selectin) ELISA Kit from Innovative Research is intended for quantitative detection of rat soluble E-Selectin in cell culture supernates, serum and plasma (heparin, citrate). Strip well format. Reagents for up to 96 tests.

This rat E-Selectin ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for E-Selectin has been precoated onto 96-well plates. Standards(NSO, W22-P494) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for E-Selectin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the rat E-Selectin amount of sample captured in plate.

Expression system for standard: E-selectin, also called endothelial-leukocyte adhesion molecule-1(ELAM-1), is a cell surface glycoprotein expressed by cytokine-activated endothelium that mediates the adhesion of blood neutrophils.1 An increased expression of E-selectin has been observed in the arterial endothelium interacting with lymphocytes and macrophages in human atherosclerotic lesions.2 E-selectin plays a critical role in mediating tissue-specific homing of T cells into skin, and of primitive hematopoietic progenitor cells(HPCs) into bone marrow(BM).3 A structurally and functionally related group of genes, lymph node homing receptor(LHR), granule membrane protein 140(GMP-140), and endothelial leukocyte adhesion molecule 1(ELAM-1) are shown to constitute a gene cluster on mouse and human chromosome 1.4; Immunogen sequence: 369
