This Mouse Thrombopoietin Receptor Sandwich ELISA Kit from Innovative Research is intended for quantitative detection of mouse TPOR in cell culture supernates, cell lysates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.
This mouse TPOR Sandwich ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for TPOR has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: Q26-W482) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for TPOR is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse TPOR amount of sample captured in plate.

The capture antibody is a monoclonal antibody from rat, the detection antibody is a biotinylated polyclonal antibody from goat. Expression system for standard: The thrombopoietin receptor (TPOR), also known as CD110, is a protein that in humans is encoded by the MPL (myeloproliferative leukemia virus) oncogene. The human MPL gene is mapped to chromosome 1p34. The protein encoded by the c-mpl gene, CD110, is a 635 amino acid transmembrane domain, with two extracellular cytokine receptor domains and two intracellular cytokine receptor box motifs. TPO-R deficient mice were severely thrombocytopenic, emphasizing the important role of CD110 and thrombopoietin in megakaryocyte and platelet formation. Upon binding of thrombopoietin CD110 is dimerized and the JAK family of non-receptor tyrosine kinases, as well as the STAT family, the MAPK family, the adaptor protein Shc and the receptors themselves become tyrosine phosphorylated.; Immunogen sequence: 369Click here to view a video tutorial covering frequently asked questions regarding ELISA workflows.
