This mouse Podocalyxin ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for Podocalyxin has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: H22-S404) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Podocalyxin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse Podocalyxin amount of sample captured in plate.

Expression system for standard: Podocalyxin-like protein 1 is a protein that in humans is encoded by the PODXL gene. It is mapped to 6 A3.3; 6 12.57 Cm. This gene encodes a member of the CD34 sialomucin protein family. The encoded protein was originally identified as an important component of glomerular podocytes. Inactivation of the encoding gene in mice leads to anuria, omphalocele and perinatal death. Podocytes are highly differentiated epithelial cells with interdigitating foot processes covering the outer aspect of the glomerular basement membrane. Other biological activities of the encoded protein include: binding in a membrane protein complex with Na+/H+ exchanger regulatory factor to intracellular cytoskeletal elements, playing a role in hematopoetic cell differentiation, and being expressed in vascular endothelium cells and binding to L-selectin.; Immunogen sequence: 369
