This mouse KLKB1 ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for KLKB1 has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: G20-A638) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for KLKB1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse KLKB1 amount of sample captured in plate.

Expression system for standard: Plasma kallikrein is a protein that in humans is encoded by the KLKB1 gene. It is mapped to 4q35.2. This gene encodes a glycoprotein that participates in the surface-dependent activation of blood coagulation, fibrinolysis, kinin generation and inflammation. The encoded preproprotein present in plasma as a non-covalent complex with high molecular weight kininogen undergoes proteolytic processing mediated by activated coagulation factor XII to generate a disulfide-linked, heterodimeric serine protease comprised of heavy and light chains. Certain mutations in this gene cause prekallikrein deficiency. Alternative splicing results in multiple transcript variants encoding different isoforms. ; Immunogen sequence: 369
