This mouse DMP-1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for DMP-1 has been precoated onto 96-well plates. Standards(NSO, L17-Y503) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for DMP-1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse DMP-1 amount of sample captured in plate.

Expression system for standard: Dentin matrix acidic phosphorprotein 1 is a protein that in humans is encoded by the DMP1 gene. It belongs to the small integrin-binding ligand N-linked glycoprotein(SIBLING) family of secreted phosphoproteins. This gene is mapped to 4q22.1. This protein, which is critical for proper mineralization of bone and dentin, is present in diverse cells of bone and tooth tissues. In undifferentiated osteoblasts, DMP-1 is primarily a nuclear protein that regulates the expression of osteoblast-specific genes. During osteoblast maturation, the protein becomes phosphorylated and is exported to the extracellular matrix, where it orchestrates mineralized matrix formation. Mutations in the gene are known to cause autosomal recessive hypophosphatemia, a disease that manifests as rickets and osteomalacia.
