This Human WNT Inducible Signaling Pathway Protein 1 Sandwich ELISA Kit from Innovative Research is intended for quantitative detection of human WISP1 in cell culture supernates, cell lysates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.
This human WISP1 Sandwich ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for WISP1 has been precoated onto 96-well plates. Standards(NSO, T23-N367) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for WISP1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human WISP1 amount of sample captured in plate.

The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. Expression system for standard: WISP1(WNT1-Inducible Signaling Pathway Protein 1), also known as CCN4, is a matricellular protein that in humans is encoded by the WISP1 gene. WISP1 is induced by WNT1 and belongs to the CCN family, which includes connective tissue growth factor, cysteine-rich-61, and nephroblastoma overexpressed. By use of radiation hybrid mapping panels, the WISP1 was mapped to chromosome 8q24.1-q24.3, roughly 4 Mb distal to MYC. It found that 2 distinct systems demonstrated that WISP induction was associated with expression of WNT1. WISP1 genomic DNA was amplified in colon cancer cell lines and in human colon tumors, and its RNA was overexpressed in 84% of tumors examined compared with patient-matched normal mucosa.; Immunogen sequence: 369Click here to view a video tutorial covering frequently asked questions regarding ELISA workflows.
