This Human Tumor Necrosis Factor Ligand Superfamily Member 12 (TWEAK) ELISA Kit from Innovative Research is intended for quantitative detection of human TNFSF12 in cell culture supernates, cell lysates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.

This human TNFSF12 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for TNFSF12 has been precoated onto 96-well plates. Standards(E.coli, S94-H249) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for TNFSF12 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human TNFSF12 amount of sample captured in plate.

Expression system for standard: Tumor necrosis factor ligand superfamily member 12 also known as TNF-related weak inducer of apoptosis(TWEAK) is a protein that in humans is encoded by the TNFSF12 gene. The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor(TNF) ligand family. This protein is a ligand for the FN14/TWEAKR receptor. This cytokine has overlapping signaling functions with TNF, but displays a much wider tissue distribution. This cytokine can induce apoptosis via multiple pathways of cell death in a cell type-specific manner. This cytokine is also found to promote proliferation and migration of endothelial cells, and thus acts as a regulator ofangiogenesis.
