This human Serpin A7 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Serpin A7 has been precoated onto 96-well plates. Standards (Expression system for standard: NSO, Immunogen sequence: A21-A415) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Serpin A7 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Serpin A7 amount of sample captured in plate.

Additional Information: The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. Expression system for standard: Thyroxine-binding globulin (TBG), also known as SERPINA7, is the major thyroid hormone transport protein in serum. There are three proteins including thyroxine-binding globulin (TBG), transthyretin and albumin responsible for carrying the thyroid hormones thyroxine (T4) and 3,5,3'-triiodothyronine (T3) in the bloodstream. This gene belongs to the serpin family in genomics, but the protein has no inhibitory function like many other members of the serpin family. Mutations in this gene result in TGB deficiency, which has been classified as partial deficiency, complete deficiency, and excess, based on the level of serum TBG. Alternatively spliced transcript variants encoding different isoforms have been found, but the full-length nature of these variants has not been determined. Using DNA blot hybridization, it is detected that there is a single-copy TBG gene located on the long arm of the X chromosome. ; Immunogen sequence: 369
