This Human Prolargin ELISA Kit from Innovative Research is intended for quantitative detection of human PRELP in cell culture supernates, serum and plasma (heparin, EDTA, citrate). Strip well format. Reagents for up to 96 tests.
This human PRELP ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for PRELP has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: Q21-I382) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for PRELP is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human PRELP amount of sample captured in plate.

The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated monoclonal antibody from goat. Expression system for standard: Prolargin is a protein that in humans is encoded by the PRELP gene. It is mapped to 1q32.1. The protein encoded by this gene is a leucine-rich repeat protein present in connective tissue extracellular matrix. This protein functions as a molecule anchoring basement membranes to the underlying connective tissue. This protein has been shown to bind type I collagen to basement membranes and type II collagen to cartilage. It also binds the basement membrane heparan sulfate proteoglycan perlecan. This protein is suggested to be involved in the pathogenesis of Hutchinson-Gilford progeria (HGP), which is reported to lack the binding of collagen in basement membranes and cartilage.
