This Human Platelet-Derived Growth Factor Receptor Alpha (CD140a) ELISA Kit from Innovative Research is intended for quantitative detection of Human PDGF-AA in cell culture supernates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.
This human PDGF-AA ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for PDGF-AA has been precoated onto 96-well plates. Standards (Expression system for standard: E.Coli, Immunogen sequence: S87-T211? and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for PDGF-AA is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human PDGF-AA amount of sample captured in plate.

Expression system for standard: PDGF-AA encodes a member of the protein family comprised of both platelet-derived growth factors (PDGF) and vascular endothelial growth factors (VEGF). The encoded preproprotein is proteolytically processed to generate platelet-derived growth factor subunit A, which can homodimerize, or alternatively, heterodimerize with the related platelet-derived growth factor subunit B. These proteins bind and activate PDGF receptor tyrosine kinases, which play a role in a wide range of developmental processes.
