This Human Platelet-Derived Growth Factor B ELISA Kit from Innovative Research is intended for quantitative detection of human PDGF-BB in cell culture supernates, cell lysates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.

This human PDGF-BB ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for PDGF-BB has been precoated onto 96-well plates. Standards(E.coli, S82-T190) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for PDGF-BB is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human PDGF-BB amount of sample captured in plate.

Expression system for standard: Platelet-derived growth factor subunit B is a protein that in humans is encoded by the PDGFB gene. The protein encoded by this gene is a member of the platelet-derived growth factor family. This gene product can exist either as a homodimer(PDGF-BB) or as a heterodimer with the platelet-derived growth factor alpha polypeptide(PDGF-AB), where the dimers are connected by disulfide bonds. This gene is mapped to 22q13.1. Growth factor plays an essential role in the regulation of embryonic development, cell proliferation, cell migration, survival and chemotaxis.
