This Human Growth Hormone Receptor ELISA Kit from Innovative Research is intended for quantitative detection of human GHR in cell culture supernates, cell lysates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.

This human GHR ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for GHR has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: A27-Y264) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for GHR is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human GHR amount of sample captured in plate.

Expression system for standard: Growth hormone receptor is a protein that in humans is encoded by the GHR gene. It is mapped to chromosome 5p13.1-p12. This gene encodes a protein that is a transmembrane receptor for growth hormone. Binding of growth hormone to the receptor leads to receptor dimerization (the receptor may however also exist as a pre-assembled non-functional dimer) and the activation of an intra- and intercellular signal transduction pathway leading to growth. Mutations in this gene have been associated with Laron syndrome, also known as the growth hormone insensitivity syndrome (GHIS), a disorder characterized by short stature (proportional dwarfism). ; Immunogen sequence: 369
