This Human Caspase 8 ELISA Kit from Innovative Research is intended for quantitative detection of human Caspase 8 in cell culture supernatesand serum. Strip well format. Reagents for up to 96 tests.

This human Caspase 8 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A polyclonal antibody from goat specific for Caspase 8 has been precoated onto 96-well plates. Standards(Expression system for standard: E. coli, Immunogen sequence: S217-D384 & L385-D479) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Caspase 8 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Caspase 8 amount of sample captured in plate.

Additional Information: The capture antibody is a polyclonal antibody from goat, the detection antibody is a biotinylated polyclonal antibody from goat. Expression system for standard: Caspase-8 is a caspase protein, encoded by the CASP8 gene. It is mapped to human chromosome 2q33-q34 and mouse chromosome 1B-proximal C. This gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes composed of a prodomain, a large protease subunit, and a small protease subunit. Activation of caspases requires proteolytic processing at conserved internal aspartic residues to generate a heterodimeric enzyme consisting of the large and small subunits. This protein is involved in the programmed cell death induced by Fas and various apoptotic stimuli. The N-terminal FADD-like death effector domain of this protein suggests that it may interact with Fas-interacting protein FADD. And this protein was detected in the insoluble fraction of the affected brain region from Huntington disease patients but not in those from normal controls, which implicated the role in neurodegenerative diseases. Many alternatively spliced transcript variants encoding different isoforms have been described, although not all variants have had their full-length sequences determined.; Immunogen sequence: 369
