This Human Angiotensinogen ELISA Kit from Innovative Research is intended for Quantitative Detection of human AGT in cell culture supernates, serum and plasma (EDTA). Strip well format. Reagents for up to 96 tests.

This human AGT ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for AGT has been precoated onto 96-well plates. Standards (Expression system for standard: NSO, Immunogen sequence: D34-A485) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for AGT is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human AGT amount of sample captured in plate.

Expression system for standard: AGT(ANGIOTENSINOGEN), also called SERPINA8, is an ?-2-globulin that is produced constitutively and released into the circulation mainly by the liver. ATG is a member of the serpin family, although it is not known to inhibit other enzymes, unlike most serpins. Angiotensinogen is also known as renin substrate. The AGT gene is mapped on 1q42.2. And the human angiotensinogen gene contains 5 exons. The expression of AGT and enzymes required for its conversion to angiotensin II in human adipose tissue. Mutations in this AGT gene are associated with susceptibility to essential hypertension, and can cause renal tubular dysgenesis, a severe disorder of renal tubular development.
