This Human Aldo-Keto Reductase Family 1 Member C3 (AKR1C3) ELISA Kit from Innovative Research is intended for quantitative detection of human AKR1C3 in cell culture supernates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.

This human AKR1C3 ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for AKR1C3 has been precoated onto 96-well plates. Standards(Expression system for standard: E.coli, Immunogen sequence: M1-Y323) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for AKR1C3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human AKR1C3 amount of sample captured in plate.

Expression system for standard: Aldo-keto reductase family 1 member C3 (AKR1C3), also known as 17?-hydroxysteroid dehydrogenase type 5 (17?-HSD5, HSD17B5) is a key steroidogenic enzyme that in humans is encoded by the AKR1C3 gene. It is mapped to 10p15.1. This gene encodes a member of the aldo/keto reductase superfamily, which consists of more than 40 known enzymes and proteins. These enzymes catalyze the conversion of aldehydes and ketones to their corresponding alcohols by utilizing NADH and/or NADPH as cofactors. The enzymes display overlapping but distinct substrate specificity. This enzyme catalyzes the reduction of prostaglandin (PG) D2, PGH2 and phenanthrenequinone (PQ), and the oxidation of 9alpha,11beta-PGF2 to PGD2. It may play an important role in the pathogenesis of allergic diseases such as asthma, and may also have a role in controlling cell growth and/or differentiation. This gene shares high sequence identity with three other gene members and is clustered with those three genes at chromosome 10p15-p14.
