Human Serum Amyloid A Chemiluminescent Immunoassay Kit from Innovative Research is a highly sensitive Chemiluminescent Immunoassay for meauring Serum Amyloid A in biofluid samples, such as serum, plasma and other biological fluids. Reagents for up to 96 tests. This assay recognizes Human SAA in samples. No significant cross-reactivity or interference between Human SAA and analogues was observed. Innovative Research's Serum Amyloid A Chemiluminescent Immunoassay kit uses the Sandwich-Chemiluminescent Immunoassay principle. The micro Chemiluminescent Immunoassay plate provided in this kit has been pre-coated with an antibody specific to Human SAA. Standards or samples are added to the micro Chemiluminescent Immunoassay plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human SAA and Avidin-Horseradish Peroxidase(HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human SAA, biotinylated detection antibody and Avidin-HRP conjugate will appear fluorescence. The Relative light unit(RLU) value is measured by the Chemiluminescence immunoassay analyzer. The RLU value is positively associated with the concentration of Human SAA. You can calculate the concentration of Human SAA in the samples by comparing the RLU value of the samples to the standard curve. • Preheat a water bath to 37 C.
• Place the frozen plasma sample directly in the water bath.
• Gently agitate the plasma sample every 5-10 minutes to insure even thawing.
• The total thawing time will depend on the frozen sample volume. A 10 ml sample might be thawed completely in 5 minutes, but a large sample could take up to 1 hour.
• The thawed sample should be used immediately or stored at 4 C for up to 24 hours. Liquid plasma should never be kept on ice or stored below 0 C as the cold may cause clotting.
• Plasma should be visually inspected for signs of clotting or precipitation before use in experiments. A coagulation test such as aPTT can also be performed to verify the preservation of the hemostasis system.
• Plasma should always be snap-frozen by placing the tube in dry ice for at least 1 hour. Large containers can be covered in dry ice for even freezing.
• Frozen plasma should be stored at –20 C or colder. Lower temperatures will help preserve labile plasma proteins for longer time periods.
