Innovative Research Human Hemoglobin Lyophilized is a lyophilized preparation derived from freshly isolated human red blood cells. Cells were lysed under hypotonic conditions to release hemoglobin. The lysate was stabilized, dialyzed into water, clarified by centrifugation, 0.22 μm filtered, and lyophilized for long-term storage and ease of use.

Characterization:

•        Size Confirmation by SDS-PAGE: 12% SDS-PAGE gels show bands at ~15 kDa, corresponding to the alpha and beta chains of hemoglobin under both reducing and non-reducing conditions. 

•        Western Blot Analysis: Chain identity confirmed using rabbit anti-HBA1-alpha and anti-HBA1-beta antibodies. 

•        Molecular Weight Distribution by Analytical SEC-HPLC:
Analytical size exclusion chromatography (BioCore SEC-120, UV at 214 nm) showed that 94.41% of total peak area corresponds to tetrameric hemoglobin (~64 kDa). A minor peak (5.6%) eluted at 12.84 min corresponds to potential dimers or dissociation products. There may be trace levels (<1%) of residual salts or buffer remnants in the material.

Functionality Testing:
Lyophilized material reconstituted at 1 mg/mL was subjected to sodium dithionite reduction followed by oxygen reoxidation. UV-Vis absorbance spectra confirmed redox cycling and functional oxygen binding.

Storage:
