This Mouse Low-Density Lipoprotein Receptor Sandwich ELISA Kit from Innovative Research is intended for quantitative detection of mouse LDLR in cell culture supernates, serum, plasma (heparin) and urine. Strip well format. Reagents for up to 96 tests.

This mouse LDLR Sandwich ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for LDLR has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: A22-R790) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for LDLR is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse LDLR amount of sample captured in plate.

The capture antibody is a monoclonal antibody from rat, the detection antibody is a biotinylated polyclonal antibody from goat. Expression system for standard: The Low-Density Lipoprotein (LDL) Receptor is a mosaic protein of 839 amino acids (after removal of 21-amino acid signal peptide) that mediates the endocytosis of cholesterol-rich LDL. In humans, the LDL receptor protein is encoded by the LDLR gene. It belongs to the Low density lipoprotein receptor gene family. The low density lipoprotein receptor (LDLR) gene family consists of cell surface proteins involved in receptor-mediated endocytosis of specific ligands. Low density lipoprotein (LDL) is normally bound at the cell membrane and taken into the cell ending up in lysosomes where the protein is degraded and the cholesterol is made available for repression of microsomal enzyme 3-hydroxy-3-methylglutaryl coenzyme A (HMG CoA) reductase, the rate-limiting step in cholesterol synthesis. At the same time, a reciprocal stimulation of cholesterol ester synthesis takes place. Mutations in this gene cause the autosomal dominant disorder, familial hypercholesterolemia.
