This Human Mucin 16 (CA-125) ELISA Kit from Innovative Research is intended for quantitative detection of human CA125 in cell culture supernates, serum, plasma (heparin, EDTA), saliva, urine and human milk. Strip well format. Reagents for up to 96 tests.

This human CA125 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for CA125 has been precoated onto 96-well plates. Standards(Expression system for standard: Natural protein) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CA125 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human CA125 amount of sample captured in plate.

Additional Information: The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. CA-125, also known as mucin 16 or MUC16, is a protein that in humans is encoded by the MUC16 gene. MUC16 is a member of the mucin family glycoproteins. It is a component of the ocular surface, the respiratory tract and the female reproductive tract epithelia. Since MUC16 is highly glycosylated it creates a hydrophilic environment that acts as a lubricating barrier against foreign particles and infectious agents on the apical membrane of epithelial cells. Also, the cytoplasmic tail of MUC16 has been shown to interact with cytoskeleton by binding members of the ERM protein family. The expression of mucin 16 has been shown to be altered in dry eye, cystic fibrosis, and several types of cancers.
