This Human Cathepsin H ELISA Kit from Innovative Research is intended for quantitative detection of human Cathepsin H in cell culture supernates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.

This human Cathepsin H ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Cathepsin H has been precoated onto 96-well plates. Standards(CHO,A23-V335) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Cathepsin H is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Cathepsin H amount of sample captured in plate.

Expression system for standard: Cathepsin H is a protein that in humans is encoded by the CTSH gene. The protein encoded by this gene gene is a lysosomal cysteine proteinase important in the overall degradation of lysosomal proteins. It is composed of a dimer of disulfide-linked heavy and light chains, both produced from a single protein precursor. The encoded protein, which belongs to the peptidase C1 protein family, can act both as an aminopeptidase and as an endopeptidase. Increased expression of this gene has been correlated with malignant progression of prostate tumors.
