This Human Autotaxin (Phosphodiesterase 2) ELISA Kit from Innovative Research is intended for Quantitative Detection of human Autotaxin in cell culture supernates, serum, plasma (heparin), urine and human milk. Strip well format. Reagents for up to 96 tests.

This human Autotaxin ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Autotaxin has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: A36-I863) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Autotaxin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Autotaxin amount of sample captured in plate.

Expression system for standard: Autotaxin, also known as ectonucleotide pyrophosphatase/phosphodiesterase family member 2 (E-NPP 2), is an enzyme that in humans is encoded by the ENPP2 gene. The protein encoded by this gene functions as both a phosphodiesterase, which cleaves phosphodiester bonds at the 5' end of oligonucleotides, and a phospholipase, which catalyzes production of lysophosphatidic acid (LPA) in extracellular fluids. LPA evokes growth factor-like responses including stimulation of cell proliferation and chemotaxis. This gene product stimulates the motility of tumor cells and has angiogenic properties, and its expression is upregulated in several kinds of carcinomas. The gene product is secreted and further processed to make the biologically active form.
