A monoclonal antibody from rat specific for TPOR has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: Q26-W482) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for TPOR is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. Expression system for standard: The thrombopoietin receptor (TPOR), also known as CD110, is a protein that in humans is encoded by the MPL (myeloproliferative leukemia virus) oncogene. The human MPL gene is mapped to chromosome 1p34. The protein encoded by the c-mpl gene, CD110, is a 635 amino acid transmembrane domain, with two extracellular cytokine receptor domains and two intracellular cytokine receptor box motifs. TPO-R deficient mice were severely thrombocytopenic, emphasizing the important role of CD110 and thrombopoietin in megakaryocyte and platelet formation.
