Upon proteolytic activation, FXI is converted to its active serine protease form, factor XIa (FXIa). FXI can be activated by factor XIIa (FXIIa) through the contact pathway and by thrombin-mediated feedback activation, resulting in amplification of coagulation through the activation of factor IX. Activated factor IX, together with factor VIIIa, promotes thrombin generation and subsequent fibrin formation. Because FXIa is a key mediator of coagulation amplification, measurement of circulating FXIa provides a valuable tool for evaluating activation of the intrinsic coagulation pathway. Enzyme-linked immunosorbent assays (ELISAs) have been developed for the sensitive detection and quantification of FXIa in biological samples.\u003c\/p\u003e\n\u003cp\u003e\u003cstrong\u003eAssay Principle\u003c\/strong\u003e\u003cbr\u003eFunctionally active FXIa will form a covalent complex with biotinylated human PAI-1 which is bound to the avidin coated on the microtiter plate. FXI will not bind to the PAI-1 and will not be detected by the assay. After appropriate washing steps, anti-human XIa primary antibody binds to the captured FXIa. Excess antibody is washed away, and bound antibody is reacted with the secondary antibody conjugated to horseradish peroxidase. Following an additional washing step, TMB is used for color development at 450nm. A standard calibration curve is prepared along with the samples to be measured using dilutions of human FXIa. The amount of color development is directly proportional to the concentration of FXIa in the sample.\u003c\/p\u003e\n\u003cp\u003eThe assay measures FXIa in the range stated below.
