This human HYAL1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for HYAL1 has been precoated onto 96-well plates. Standards (Expression system for standard: NSO, Immunogen sequence: F22-W435) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for HYAL1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human HYAL1 amount of sample captured in plate.

Expression system for standard: Hyaluronidase-1, also known as HYAL1 or LUCA1, is an enzyme that in humans is encoded by the HYAL1 gene. The gene is one of several related genes in a region of chromosome 3p21.3 associated with tumor suppression. This gene encodes a lysosomal hyaluronidase. Hyaluronidases intracellularly degrade hyaluronan, one of the major glycosaminoglycans of the extracellular matrix. Hyaluronan is thought to be involved in cell proliferation, migration and differentiation. This enzyme is active at an acidic pH and is the major hyaluronidase in plasma.
